Background and overview[1]
Vitamin phenol is a synthetic product of vitamins and phenol and can be used as a pharmaceutical synthesis intermediate. If vitamin phenol is inhaled, move the patient to fresh air; if skin contact occurs, take off contaminated clothing, rinse the skin thoroughly with soap and water, and seek medical attention if you feel uncomfortable; if contact with eyes, separate eyelids, Rinse with running water or normal saline and seek medical attention immediately. If ingested, rinse mouth immediately. Do not induce vomiting and seek medical attention immediately.
Preparation[1]
Vitaminol was prepared as follows: To a suspension of N-hydroxysuccinimidyl biotin (6.2 mg, 18.2 μmol) in 500 μL methanol was added 1-propanolamine (1.39 μL, 20.0 μmol). After stirring for 24 hours at 37°C, the resulting solution was purified by reverse-phase HPLC (dp5μ, 10mm × 25cm, 2mL/min, 40 to 75% acetonitrile/water in 15 minutes, retention time: 12.1 minutes) to give vitamin phenol (4.8mg, 88%): 1HNMR (500MHz, CD3OD) δ4.49 (dd, J=8.0, 5.0Hz, 1H), 4.30 (ddJ=8.0, 5.0Hz, 1H), 3.57 (t, J=6.5Hz , 2H), 3.35 (t, J=7.0Hz, 2H), 3.31 (m, 1H), 3.02 (dd, J=13.0, 5.0Hz, 1H), 2.79 (d, J=12.5Hz, 1H), 2.30 (t, J=7.5Hz, 2H), 1.70 (m, 4H), 1.63 (m, 2H) 1.43 (m, 2H); 13CNMR (125MHz, CD3OD) δ176.3, 63.4, 61.6, 60.4, 57.0, 41.0 , 37.4, 36.8, 33.2, 29.8, 29.5, 26.9; FT-IR (thin film) 3287, 3083, 2920, 2863, 1701, 1685, 1632, 1620, 1543, 1509, 1478, 1458, 1323, 1265, 1240, 1205 , 1142, 1073, 1051cm-1C13H24N3O3S calculated HRMS (TOFES) value 302.1538m/z (M+H)+; observed 302.1528 (3.3ppm error).
