Cell Culture Grade Dimethyl Sulfoxide DMSO Product Description

DMSO (dimethyl sulfoxide) is an aprotic polar solvent and is commonly used in chemical dimethyl sulfoxide reactions, PCR reactions and in cells, tissues and organs. It is used as a vitrification cryoprotectant in preservation. DMSO is used in cell freezing media to protect cells from mechanical damage caused by ice crystals. It can also be used for cryopreservation of main culture, subculture, recombinant aneuploid, hybridoma and other series of cell lines, embryonic stem cells (ESC) and hematopoietic stem cells. In addition, it is often mixed with BSA or FBS.

Uses of cell culture grade dimethyl sulfoxide DMSO

(1) DMSO is widely used in the cryogenic protection of human and animal cell lines and bacterial phage lambda. The steps for preparing DMSO solution for freezing cells are as follows:
1) Prepare freezing medium: containing cell culture medium, 10-20% serum and cerium carbonate 5-10% DMSO;
2) Use trypsin or other appropriate methods to digest adherent cells; ( Cells are best in the logarithmic growth phase)
3) Collect the cells by low-speed centrifugation (4 °C, 250 × g, 10min), and aspirate the culture medium;
4) Suspend the cells in freezing culture medium, and The cell density is 106 -107 cells/ml;
5) Evenly distribute the cells and put them into cryovials;
6) Freeze the cells according to standard freezing procedures and freeze them at -70 °C or lower within the temperature;
(2) When cells are fused, add 10% DMSO to the 40-50% PEG solution;
(3) There are reports in the literature that DMSO is used as a component of the buffer and is used in the following experiments: Poly (A+) RNA purification, E. coli competent cell transformation, PCR reaction, cDNA library amplification, DNA sequencing, DEAE-dextran-mediated cell transfection, polybrene-mediated DNA transfection;
(4) There are corresponding procedures describing the use of DMSO to recover membrane-filtered DNA for subsequent PCR amplification reactions;
(5) Capillary electrophoresis technology used for DNA sequencing converts 5% DMSO (v/w) mixed into 2 M urea and adjust DMSO concentration to 100% if necessary.
(6) Some studies have reported the relationship between different concentrations of DMSO and the melting point temperature of oligonucleotides;
(7) Some studies have described the use of DMSO to increase the production of monoclonal antibodies in hybridoma cells; Investigations have found that low temperature Hybridoma cells were cultured in DMSO-containing medium at high temperature before freezing.

Instructions for use of cell culture grade dimethyl sulfoxide DMSO

DMSO mixes easily with water (1 ml DMSO + 1 ml H2O) to form a clear, colorless solution. Easy to absorb moisture, avoid exposure to humid environment. Also soluble in ethanol, acetone, ether, benzene, and chloroform;

Cell culture grade DMSO storage/stability

DMSO is easily supercooled and melts slowly at room temperature. The product received should be solid and not liquid. Solidified DMSO can be liquefied again when placed at room temperature without damaging itself. It is a thermally stable compound and remains stable in acids, bases and neutral solutions up to 100 °C. The temperature is close to its boiling point (189°C) and remains stable in alkaline and neutral solutions. When heated at 150°C for 24 hours, the purity loss is less than 0.1%.
To prepare a DMSO solution for membrane sterilization, it is recommended to use teflon or nylon membrane, and try not to use cellulose acetate membrane.
